STUDY OF MICROTUBERIZATION DYNAMICS OF POTATO (SOLANUM TUBEROSUM L.) CV. NEVSKII IN VITRO

Summary. The investigations were carried out in order to study the dynamics of microtuberization of potato (Solanum tuberosum L.) cv. Nevskii in vitro culture under influence of different factors. Plants were cultivated on Murashige and Skoog medium. We studied the influence of sucrose (3 (control), 5, 6 and 8%) under conditions of short day (8/16 h, SD) and in the dark (0/24 h, D), as well as the effect of kinetin (1, 1.75, 2.5 mg/l) and benzylaminopurine (1, 3, 5, 8 mg/l) against the background of 8% sucrose (control) under the same lighting conditions. The terms of tuberization and the relative number of productive plants (RNPP) was assessed during 12 weeks. In the dark on hormone-free culture medium microtubers appeared in 6 weeks, and with the short day – in 9 weeks of incubation. On hormone-free culture medium, photoperiod was the main factor, determining the time of tuber initiation. Under SD conditions on the culture medium, containing kinetin (1 mg/l) and benzylaminopurine (1 mg/l), microtubers appeared during the first week of incubation. In the dark on kinetin culture media (1, 1.75, 2.5 mg/l) microtubers appeared in the second week, on benzylaminopurine media (3 mg/l) – in the third week. Against the background of the optimal concentration of sucrose (8%) exogenous cytokinins (kinetin and benzylaminopurine) were the main factor determining tuber initiation time. Against 8% sucrose in the dark RNPP was 45% (p = 0.014). Under D and SD conditions with kinetin added RNPP reached 80-90% (p is less 0.05); in the dark with 5 mg/l benzylaminopurine – 78% (p = 0.04). For the reduction of microtuber induction term and increase in RNPP the combination of kinetin (1, 1.75, 2.5 mg/l) or benzylaminopurine (5 mg/l) with the optimal concentration of sucrose (8%) under SD or D conditions is required.
