Effects of sucrose, kinetin, benzoaminopurine, and photoperiod on the weight and number of microtubers in aseptic in vitro culture of Nevsky potato (Solanum tuberosum L.) variety

Abstract. The studies aimed to optimize the conditions for obtaining potato microtubers in aseptic culture in vitro. We studied the effect of sucrose, cytokinins, and photoperiod on the formation of microtubers on single-nodal microtubules of Nevsky potato (Solanum tuberosum L.) variety. The plants were cultivated on the Murashige-Skoog (MS) nutrient medium. In the first series of experiments, we compared the effects of different sucrose (Suc) rates (30 g/L (control), 50 g/L, 60 g/L, and 80 g/L) on in vitro tuberization on agar medium MS under conditions of a short day (8/16 h, SD) and continuous darkness (0/24 h, CD). In the second series of experiments, we assessed the effect of different kinetin (Kin) (1 mg/L, 1.75 mg/L, and 2.5 mg/L) and benzoaminopurine (BAP) (1 mg/L, 3 mg/L, 5 mg/L, and 8 mg/L) rates at the sucrose rate of 80 g/L (control) under SD and CD conditions. Microplants were incubated for 12 weeks. For a comprehensive assessment of the experimental options, we used an integrated indicator of the average productivity of microplants (APM). The mass of microtubers under SD conditions on the nutrient medium containing sucrose varied from 6 to 361 mg; on the medium containing phytohormones, it varied from 10 to 618 mg; under CD conditions, it varied from 3 to 36 mg and from 3 to 313 mg, respectively. Variance analysis revealed a significant effect of the photoperiod on the mass of microtubers. Exogenous phytohormones influenced the mass of microtubers only under CD conditions. Significant differences with the control were noted in the options with nutrient media Kin1-Suc 80 (p = 0.002), Kin1.75-Suc80 (p = 0.008), Kin2.5-Suc80 (p = 0.005), and BAP5-Suc80 (p = 0.047). Under SD conditions, indicators in the control and cytokinin options were the same. In terms of APM, the culture medium Suc80 (122.3 mg/plant) distinguished itself under SD conditions (p = 0.002).
