ESTIMATION OF QUANTITATIVE CHARACTERISTICS OF POTATO LEAF HAIRINESS USING MICROSCOPIC IMAGE ANALYSIS

Summary. Quantitative characteristics of leaf indumentum for 35 varieties of potato were estimated using analysis of microscopic images. The plants were grown in a field of the Siberian Research Institute for Plant Industry and Breeding (Novosibirsk region) from late May to early August 2016. Leaf samples were collected and processed from 3 to 5 August 2016. Fragments of leaves of 1.5 x 0.5 cm were taken between the veins, cut in the laboratory and fold across, the bottom surface of the leaf inside. Fixation of the leaf fragment on a slide was made using sticky tape with the 3 mm offset from the rim of the fold. The glass slide was placed under a digital camera Axiocam HRc of the optical microscope ZEISS Axioscop 2 with an adapter at the magnification of 0.63 in the transmitted light. The obtained images were processed by the program LHDetect2, which enables to count the number of trichoma in the image and estimate the length of each of them. The data were averaged over 6 leaf samples for one plant. For the studied varieties, the average number of trichoma in the image ranged from 1 (weakly downy varieties Krasavchik, Impala, Bryanskii, Reggi) to 7.3 (very downy varieties Kemerovchanin, Velikan, Tanai varieties). The average value for the sample was 2.94, the value of a standard deviation was 1.55. The average length of trichoma varied from 120 to 150 mcm (Krasavchik, Safo, Tango, Kemerovchanin varieties) to 380 mcm (Kortni variety). The average value for the sample was 207.39 mcm, the standard deviation was 58.30 mcm. Significant relationship between the number of trichoma and their length in plants was not detected (Pearson correlation coefficient was -0.147, p is less than 0.39).
